A 2.3 kDa of antimicrobial peptide was purified from an acidified liver extract of skipjack tuna, Katsuwonus pelamis, by preparative acid-urea-polyacrylamide gel electrophoresis and C18 reversed-phase HPLC. A comparison of the amino acid sequence of the purified peptide with those of other known polypeptides revealed high homology with the C-terminus of hemoglobin β-chain; thus, this peptide was designated as the Skipjack Hemoglobin β chain-related Antimicrobial Peptide(SHβAP). SHβAP showed potent antimicrobial activity against Gram-positive bacteria, such as Bacillus subtilis, Staphylococcus aureus, and Streptococcus iniae (minimal effective concentrations [MECs], 6.5-57.0 Î¼g/mL), Gram-negative bacteria, such as Escherichia coli D31, Pseudomonas aeruginosa, Salmonella enterica, Shigella sonnei, and two Vibrio parahaemolyticus species (MECs, 2.0-19.0 Î¼g/mL), and against Candida albicans (MEC; 12.0 Î¼g/mL) without significant hemolytic activity. Antimicrobialactivity of this peptide was heatstable and pH resistant but is sensitive to proteases and salt. SHβAP did not show membrane permeabilization and killing ability. The secondary structural prediction and the homology modeling expected that this peptideformed an amphipathic α-helical structure. This is the first report the purification of a novel antimicrobial peptide related to the C-terminus of hemoglobin β-chain from marine fish.

If classic movies like Glengarry Glen Ross (1992), Tin Men (1987), Used Cars (1980)—or more recent hits like Wolf of Wall Street (2013)—have taught us anything, it’s that sales is not for those of a weak disposition.

And whether the character portrayal is ruthless and intimidating like Alec Baldwin’s Blake, broadly comedic like Danny DeVito’s Ernest Tilley, or charmingly sneaky like Kurt Russell’s Rudy Russo, movies have also suggested that salespeople need to be either pushy or conniving to achieve success.

While such representations do elevate dramatic tension or provide an impetus for farcical scenarios to play out on screen, they’re pretty far from reality, especially in today’s world of sales. Now more than ever, customers are informed and have a wide array of choices. Online reviews, comparison websites, specs, even satellite and street views of real estate are at the fingertips of potential buyers. So if salespeople have lost their leverage with information, what can they do to win the account?

Some attributes typically associated with sales remain important and probably will be for some time. Building relationships, for example, is something even the most advanced predictive algorithm cannot do. Persuasiveness, coupled with social engagement (which we might describe as “personal impact”), is also a major component of selling. As implied in the opening paragraph above, resilience is perhaps most critical of all to sticking it out in competitive sales. The best salesperson loses at least 75% of the time.

Alas, personal impact and resilience are not teachable qualities. There are compensatory techniques that can be conveyed through training and coaching, but both psychometric research and practical experience show that such techniques are rarely sustainable in sales. If you work against your intrinsic motivations, like cold-calling when you are socially reserved and lack self-assurance for example, you are likely to burn out sooner rather than later.

So let us assume we’re addressing salespeople who already display personal impact, are adept at relationship building, and can rebound from rejection. How can they adapt in a sales environment where information technology has empowered customers as never before? By demonstrating the fourth common success factor revealed by psychometric research: curiosity.

In measuring the personality drivers of successful sales people across multiple industries, we have found that “information seeking” is a quality shared among many top performers. We’re not talking about following a sales script or asking basic qualifying questions. Information seekers are naturally curious about people and things. Information seekers are not dismissive of new ideas and input, either, and they are open to others’ perspectives and experiences. Your grumpy uncle who reflexively thinks anything that does not interest him is a stupid waste of time? Not an information seeker. Maybe he did great in insurance sales thirty or forty years ago, but he’d find a very different environment today.

How does information-seeking lead to sales success? Now that’s a question for curious people! The folks who bailed on this piece after reading the previous paragraph—because they reflexively dismissed the concept—were not information seekers either. Let’s explore the answer without them.

Information seeking supports sales success because it redirects the conversation. The customer did the research and already has the specs on hand and prices in mind. They think they are in control. When you start asking questions—meaningful, probing questions they weren’t expecting—the information you elicit gets them thinking and rethinking, perhaps about potentialities they overlooked. You’re showing them that you have a whole level of understanding they hadn’t imagined, giving you instant credibility. Now they’re more receptive to the message you intended to convey all along.

Information seeking doesn’t always manifest in the form of overt questions. It could be quiet observation, or it could be collecting seemingly irrelevant data that you process in the back of your mind. It could be simple environmental scanning. Salespeople who are open to such inputs are the ones who, once the clues coalesce in their minds, find the right button to push or notice hidden opportunities that turn a minor sale into a major one.

Contrast that with the “old-school” sales approach, in which the sales representative already has his mind made up about what clients want and need and shuts down any chance of engagement. He might be friendly, persuasive, and resilient to setbacks, but his close-minded, inflexible approach leaves money on the table.

Curiosity in sales isn’t a new thing, of course, but simply an attribute that hasn’t always been considered in the applicant-selection process. Recent performance data says it should be. So consider stirring in a little Sherlock Homes with your Glengarry Glen Ross. Or should we call it, perhaps, The Case of the Missing Leads?

The Methods of Using the Extruder Screw Elements

The daily maintenance of the screw barrel is a problem that many people in the industry are concerned about, so what are the specifics?

Josh Mart takes the chance to preview the second leg of this year’s UEFA Champions League round of 16 between FC Barcelona and Paris Saint Germain at the Camp Nou in Barcelona.

Since the heavy defeat in Paris, Barcelona have been near perfect domestically scoring a mightily impressive 15th goal in their last 4 games. Saturday’s 5-0 thrashing of Celta Vigo was considered by some to be Barcelona’s most complete performance of the season.

This coincides with Luis Enrique’s decision to switch to a 3-4-3 formation which could be deployed against PSG tonight. Coach Luis Enrique has recently announced that he will be leaving Barcelona at the end of the season, but he and his squad will be desperate to make history and pull of the near impossible at the Camp Nou this evening.

PSG have also been in great form in France, despite a minor European hangover after the Barcelona rout where they drew 0-0 at home to Toulouse. A fabulous 5-1 victory in ‘Les Clasique’ away to Marseille has been followed up by a Coupe de France win over Niort and a routine 1-0 victory over Nancy at the Parc des Princes on Saturday.

Despite their good form, Unai Emery’s men still behind leaders Monaco in the Ligue 1 standings but their spectacular performance against Barcelona has gone some way to silencing Emery’s critics this season.

Team News and Tactics:

Barcelona are without the injured duo of Aleix Vidal and Jeremie Mathieu for this one, but Luis Enrique’s main decision to make is whether to play the 3-4-3 formation which worked so well against Celta at the weekend, or revert to the clubs typical 4-3-3 formation.

I think that Barcelona will line up with the 3-4-3 with Ter Stegen behind a back 3 of Mascherano, Pique and Umtiti. The midfield will be quite narrow to try and gain more control of the ball- something which they lacked terribly at the Parc des Princes. Sergio Busquets and Andres Iniesta look set to start and will most probably be joined by Ivan Rakitic and one of Jordi Alba, Sergi Roberto or Andre Gomes depending on how much Enrique wants from the midfield. The front 3 of Messi, Suarez and Neymar picks itself, despite a very under par performance from them all 3 weeks ago.

Unai Emery really does have some big selection problems here, as he is missing Thiago Motta and probably Di Maria through injury and there has been speculation about Marco Verratti and Adrien Rabiot missing out too.

With this in mind, I expect Unai Emery to stick with his 4-3-3 formation with Trapp in goals behind a defence of Meunier, Marquinhos, Thiago Silva and Kurzawa. Kimpembe could retain his place but the experience of Thiago Silva may be needed here. If the injury rumours are to be believed it would be very tricky to assume PSG’s midfield trio, but I think at least one of Verratti/Rabiot will play alongside Matuidi in midfield. The other option (if needed) could be filled by Krychowiak or the talented youngster Nkunku. Di Maria’s void is likely to be filled by Lucas Moura, who will play in the front three alongside Julian Draxler and Edinson Cavani.

Julian Draxler (PSG) – Julian Draxler’s career promised so much when he broke through and became an integral part of the first team at Schalke but he was criticised heavily (partly justifiably) for what was seen as a sideways move to Wolfsburg. Since moving to Paris though, he has been superb. The talent which has never been in question is now accompanied by a hunger and desire to win which was so evident in the first leg.

Prediction

This is a very strange game to predict because it is not often that there is such a large margin in the aggregate scores at this level of competition. It’s hard not to see Barcelona winning, purely because PSG have no need to try and win the match and because they will know that their pride is at stake here. I think Barcelona will win the game, but PSG will go through thanks to their marvellous first leg performance.

Jamie Vardy insulted Samir Nasri’s family, leading to the Frenchman’s sending off

Leicester City striker Jamie Vardy allegedly insulted Sevilla’s French attacking midfielder Samir Nasri’s family last night, leading to the Manchester City loanee to react in a movement that was interpreted as a head-butt, according to France Football.
